Disease-associated mutations in KCNE potassium channel subunits (MiRPs) reveal promiscuous disruption of multiple currents and conservation of mechanism.
KCNE genes encode single transmembrane-domain subunits, the MinK-related peptides (MiRPs), which assemble with pore-forming alpha subunits to establish the attributes of potassium channels in vivo. To investigate whether MinK, MiRP1, and MiRP2 operate similarly with their known native alpha subunit partners (KCNQ1, HERG, and Kv3.4, respectively) two conserved residues associated with human disease and influential in channel function were evaluated. As MiRPs assemble with a variety of alpha subunits in experimental cells and may do so in vivo, each peptide was also assessed with the other two alpha subunits. Inherited mutation of aspartate to asparagine (D --> N) to yield D76N-MinK is linked to cardiac arrhythmia and deafness; the analogs D82N-MiRP1 and D90N-MiRP2 were studied. Mutation of arginine to histidine (R --> H) to yield R83H-MiRP2 is associated with periodic paralysis; the analogs K69H-MinK and K75H-MiRP1 were also studied. Macroscopic and single-channel currents showed that D --> N mutations suppressed a subset of functions whereas R/K --> H changes altered the activity of MinK, MiRP1, and MiRP2 with all three alpha subunits. The findings indicate that the KCNE peptides interact similarly with different alpha subunits and suggest a hypothesis: that clinical manifestations of inherited KCNE point mutations result from disruption of multiple native currents via promiscuous interactions

Phosphorylation and protonation of neighboring MiRP2 sites: function and pathophysiology of MiRP2-Kv3.4 potassium channels in periodic paralysis.
MinK-related peptide 2 (MiRP2) and Kv3.4 subunits assemble in skeletal muscle to create subthreshold, voltage-gated potassium channels. MiRP2 acts on Kv3.4 to shift the voltage dependence of activation, speed recovery from inactivation, suppress cumulative inactivation and increase unitary conductance. We previously found an R83H missense mutation in MiRP2 that segregated with periodic paralysis in two families and diminished the effects of MiRP2 on Kv3.4. Here we show that MiRP2 has a single, functional PKC phosphorylation site at serine 82 and that normal MiRP2-Kv3.4 function requires phosphorylation of the site. The R83H variant does not prevent PKC phosphorylation of neighboring S82; rather, the change shifts the voltage dependence of activation and endows MiRP2-Kv3.4 channels with sensitivity to changes in intracellular pH across the physiological range. Thus, current passed by single R83H channels decreases as internal pH is lowered (pK(a) approximately 7.3, consistent with histidine protonation) whereas wild-type channels are largely insensitive. These findings identify a key regulatory domain in MiRP2 and suggest a mechanistic link between acidosis and episodes of periodic paralysis
Kcne4 deletion sex- and age-specifically impairs cardiac repolarization in mice.
Myocardial repolarization capacity varies with sex, age, and pathology; the molecular basis for this variation is incompletely understood. Here, we show that the transcript for KCNE4, a voltage-gated potassium (Kv) channel β subunit associated with human atrial fibrillation, was 8-fold more highly expressed in the male left ventricle compared with females in young adult C57BL/6 mice (P < 0.05). Similarly, Kv current density was 25% greater in ventricular myocytes from young adult males (P < 0.05). Germ-line Kcne4 deletion eliminated the sex-specific Kv current disparity by diminishing ventricular fast transient outward current (Ito,f) and slowly activating K(+) current (IK,slow1). Kcne4 deletion also reduced Kv currents in male mouse atrial myocytes, by >45% (P < 0.001). As we previously found for Kv4.2 (which generates mouse Ito,f), heterologously expressed KCNE4 functionally regulated Kv1.5 (the Kv ι subunit that generates IKslow1 in mice). Of note, in postmenopausal female mice, ventricular repolarization was impaired by Kcne4 deletion, and ventricular Kcne4 expression increased to match that of males. Moreover, castration diminished male ventricular Kcne4 expression 2.8-fold, whereas 5ι-dihydrotestosterone (DHT) implants in castrated mice increased Kcne4 expression >3-fold (P = 0.01) to match noncastrated levels. KCNE4 is thereby shown to be a DHT-regulated determinant of cardiac excitability and a molecular substrate for sex- and age-dependent cardiac arrhythmogenesis

Hypothalamic actions of neuromedin U.
The central nervous system and gut peptide neuromedin U (NMU) inhibits feeding after intracerebroventricular injection. This study explored the hypothalamic actions of NMU on feeding and the hypothalamo-pituitary-adrenal axis. Intraparaventricular nucleus (intra-PVN) NMU dose-dependently inhibited food intake, with a minimum effective dose of 0.1 nmol and a robust effect at 0.3 nmol. Feeding inhibition was mapped by NMU injection into eight hypothalamic areas. NMU (0.3 nmol) inhibited food intake in the PVN (0-1 h, 59 Âą 6.9% of the control value; P < 0.001) and arcuate nucleus (0-1 h, 76 Âą 10.4% of the control value; P < 0.05). Intra-PVN NMU markedly increased grooming and locomotor behavior and dose-dependently increased plasma ACTH (0.3 nmol NMU, 24.8 Âą 1.9 pg/ml; saline, 11.4 Âą 1.0; P < 0.001) and corticosterone (0.3 nmol NMU, 275.4 Âą 40.5 ng/ml; saline, 129.4 Âą 25.0; P < 0.01). Using hypothalamic explants in vitro, NMU stimulated CRH (100 nM NMU, 5.9 Âą 0.95 pmol/explant; basal, 3.8 Âą 0.39; P < 0.01) and arginine vasopressin release (100 nM NMU, 124.5 Âą 21.8 fmol/explant; basal, 74.5 Âą 7.6; P < 0.01). Leptin stimulated NMU release (141.9 Âą 20.4 fmol/explant; basal, 92.9 Âą 9.4; P < 0.01). Thus, we describe a novel role for NMU in the PVN to stimulate the hypothalamo-pituitary-adrenal axis and locomotor and grooming behavior and to inhibit feeding
Upper limits on the observational effects of nuclear pasta in neutron stars
The effects of the existence of exotic nuclear shapes at the bottom of the
neutron star inner crust - nuclear `pasta' - on observational phenomena are
estimated by comparing the limiting cases that those phases have a vanishing
shear modulus and that they have the shear modulus of a crystalline solid . We
estimate the effect on torsional crustal vibrations and on the maximum
quadrupole ellipticity sustainable by the crust. The crust composition and
transition densities are calculated consistently with the global properties,
using a liquid drop model with a bulk nuclear equation of state (EoS) which
allows a systematic variation of the nuclear symmetry energy. The symmetry
energy J and its density dependence L at nuclear saturation density are the
dominant nuclear inputs which determine the thickness of the crust, the range
of densities at which pasta might appear, as well as global properties such as
the radius and moment of inertia. We show the importance of calculating the
global neutron star properties on the same footing as the crust EoS, and
demonstrate that in the range of experimentally acceptable values of L, the
pasta phase can alter the crust frequencies by up to a factor of three,
exceeding the effects of superfluidity on the crust modes, and decrease the
maximum quadrupole ellipticity sustainable by the crust by up to an order of
magnitude. The signature of the pasta phases and the density dependence of the
symmetry energy on the potential observables highlights the possibility of
constraining the EoS of dense, neutron-rich matter and the properties of the

Remnants of Initial Anisotropic High Energy Density Domains in Nucleus-Nucleus Collisions
Anisotropic high energy density domains may be formed at early stages of
ultrarelativistic heavy ion collisions, e.g. due to phase transition dynamics
or non-equilibrium phenomena like (mini-)jets. Here we investigate hadronic
observables resulting from an initially created anisotropic high energy density
domain. Based on our studies using a transport model we find that the initial
anisotropies are reflected in the freeze-out multiplicity distribution of both
pions and kaons due to secondary hadronic rescattering. The anisotropy appears
to be stronger for particles at high transverse momenta. The overall kaon
multiplicity increases with large fluctuations of local energy densities, while
no change has been found in the pion multiplicity.Comment: Submitted to PR
Recovery of Barotrauma Injuries in Chinook Salmon, Oncorhynchus tshawytscha from Exposure to Pile Driving Sound
Juvenile Chinook salmon, Oncorhynchus tshawytscha, were exposed to simulated high intensity pile driving signals to evaluate their ability to recover from barotrauma injuries. Fish were exposed to one of two cumulative sound exposure levels for 960 pile strikes (217 or 210 dB re 1 ÂľPa2¡s SELcum; single strike sound exposure levels of 187 or 180 dB re 1 ÂľPa2â
s SELss respectively). This was followed by an immediate assessment of injuries, or assessment 2, 5, or 10 days post-exposure. There were no observed mortalities from the pile driving sound exposure. Fish exposed to 217 dB re 1 ¾Pa2¡s SELcum displayed evidence of healing from injuries as post-exposure time increased. Fish exposed to 210 dB re 1 ¾Pa2¡s SELcum sustained minimal injuries that were not significantly different from control fish at days 0, 2, and 10. The exposure to 210 dB re 1 ¾Pa2¡s SELcum replicated the findings in a previous study that defined this level as the threshold for onset of injury. Furthermore, these data support the hypothesis that one or two Mild injuries resulting from pile driving exposure are unlikely to affect the survival of the exposed animals, at least in a laboratory environment
Revisiting special relativity: A natural algebraic alternative to Minkowski spacetime
Minkowski famously introduced the concept of a space-time continuum in 1908,
merging the three dimensions of space with an imaginary time dimension , with the unit imaginary producing the correct spacetime distance , and the results of Einstein's then recently developed theory of special
relativity, thus providing an explanation for Einstein's theory in terms of the
structure of space and time. As an alternative to a planar Minkowski space-time
of two space dimensions and one time dimension, we replace the unit imaginary , with the Clifford bivector for the plane
that also squares to minus one, but which can be included without the addition
of an extra dimension, as it is an integral part of the real Cartesian plane
with the orthonormal basis and . We find that with this model of
planar spacetime, using a two-dimensional Clifford multivector, the spacetime
metric and the Lorentz transformations follow immediately as properties of the
algebra. This also leads to momentum and energy being represented as components
of a multivector and we give a new efficient derivation of Compton's scattering
formula, and a simple formulation of Dirac's and Maxwell's equations. Based on
the mathematical structure of the multivector, we produce a semi-classical
model of massive particles, which can then be viewed as the origin of the
Minkowski spacetime structure and thus a deeper explanation for relativistic
effects. We also find a new perspective on the nature of time, which is now
